美國斯隆-凱特琳研究所等處的研究人員創(chuàng)建了一種多能干細(xì)胞基因組編輯平臺：iCRISPR，這一平臺能快速，高效的敲除干細(xì)胞中的基因，而且還能在干細(xì)胞分化過程中，進(jìn)行階段特異性的基因敲除，這將在人類疾病復(fù)雜病理研究中大放異彩。相關(guān)文章發(fā)表于2014年6月12日的《Cell Stem Cell》雜志上。
人體多能干細(xì)胞(hPSCs)不僅能被用于臨床的再生研究應(yīng)用中，而且也能作為解析復(fù)雜性狀和特征的*平臺，闡明其背后的基因和分子途徑。為了實(shí)現(xiàn)這一目的，科學(xué)家們開發(fā)了多種遺傳操控方法，但是這些方法依然存在各種問題，我們需要快速，具有可操控性的生物學(xué)手段。

除了這項(xiàng)研究之外，去年來自加州大學(xué)舊金山分校的研究人員提出了一個相似的名稱：CRISPRi，他們發(fā)現(xiàn)當(dāng)缺失核酸內(nèi)切酶活性的Cas9與一種導(dǎo)向RNA共表達(dá)時候，會產(chǎn)生一種DNA識別復(fù)合物，這種復(fù)合物能特異性干擾轉(zhuǎn)錄延伸，RNA聚合酶結(jié)合，或轉(zhuǎn)錄因子結(jié)合。

加州大學(xué)圣迭戈分校(University of California, San Diego)醫(yī)學(xué)院研究人員針對自閉癥研究出一套比較新銳的理論，并對其進(jìn)行了實(shí)際測試。研究結(jié)果顯示，自閉癥實(shí)際上是由異常的細(xì)胞通訊導(dǎo)致的，使用問世已逾百年的治療嗜睡癥的藥物，可以恢復(fù)患有自閉癥的老鼠體內(nèi)的細(xì)胞活動，消除老鼠的神經(jīng)失調(diào)癥狀。參與實(shí)驗(yàn)的老鼠年齡相當(dāng)于人類30歲左右。相關(guān)研究成果在2014年6月17日的在線刊物《平移精神病學(xué)(Translational Psychiatry)》中發(fā)表。
